16, (5α)-ANDROSTEN-3α-OL (Alpha-Androstenol) is a pheromone that is produced in both men and women and is considered a social while displaying sexual aspects. This molecules fragrance has been described as musky with hints of sandalwood, or sweet vanilla (Sandalwood: woody, earthy, spicy and sometimes acrid).

Although considered mostly for social usage Alpha-Androstenol has contributed, in study, to both men and women appearing as sexier, more attractive and approachable as opposed to those without. Abstract

The orphan receptor CAR-beta binds DNA as a heterodimer with the retinoid-X receptor and activates gene transcription in a constitutive manner. Here we show that, in contrast to the classical nuclear receptors, the constitutive activity of CAR-beta results from a ligand-independent recruitment of transcriptional co-activators. While searching for potential ligands of CAR-beta, we found that the steroids androstanol and androstenol inhibit the constitutive activity of CAR-beta. This effect is stereospecific: only 3alpha-hydroxy, 5alpha-reduced androstanes are active. These androstanes do not interfere with heterodimerization or DNA binding of CAR-beta; instead, they promote co-activator release from the ligand-binding domain. These androstane ligands are examples of naturally occurring inverse agonists that reverse transcriptional activation by nuclear receptors. CAR-beta (constitutive androstane receptor-beta), therefore, defines an unanticipated steroidal signalling pathway that functions in a manner opposite to that of the conventional nuclear receptor pathways.

Abstract

The hypothesis that 5-alpha-androst-16-en-3 alpha-ol increases the sexual arousal of the human female was examined. This substance is produced by the male and has been suggested to be a possible human pheromone. Groups of female subjects were asked to read either a neutral or a sexually-arousing passage in the presence of either androstenol or a placebo. Although sexual arousal was clearly manipulated by the experimental conditions, there was no evidence that androstenol influenced sexual feelings.

Human exposure to putative pheromones and changes in aspects of social behaviour.

Abstract

Student volunteers (38 of each sex) were exposed unknowingly overnight to the vapour of pheromonally active substances and compared with controls. The substances were either 5 alpha-16-androsten-3 alpha-ol (androstenol, occurring in human underarm sweat, and known to be pheromonally active in pig and man), or a mixture of short-chain fatty acids (occurring in human vaginal fluid, and known to be sexually attractive to the male rhesus monkey). The following morning, the subjects provided information about their social exchanges since rising, by recording on a standardized test diagram the number, depth, duration and direction of initiation, of all verbal exchanges with other individuals. Irrespective of treatment, males returned significantly higher scores than did females for all exchanges and also for some exchanges initiated by other males. Neither exposure to androstenol nor to the fatty acids had any significant effects on any of the scores of males interacting with either sex, nor on any scores of females with other females. However, exposure of females to androstenol, but not to the fatty acids, resulted in significantly higher scores of exchanges with males, in terms of all parameters for all exchanges. Findings are considered in relation to the origin and maintenance across species of pheromonal communication: evolutionary conservation is seen in terms of the utilization of substances that have provided the means of controlling the social milieu.

Abstract

Androstenol is a steroidal compound belonging to the group of odorous 16-androstenes, first isolated from boar testes and also found in humans. Androstenol has pheromone-like properties in both animals and humans, but the molecular targets of its pheromonal activity are unknown. Androstenol is structurally similar to endogenous A-ring reduced neurosteroids that act as positive modulators of GABA(A) receptors. Here we show that androstenol has neurosteroid-like activity as a GABA(A) receptor modulator. In whole-cell recordings from cerebellar granule cells, androstenol (but not its 3beta-epimer) caused a concentration-dependent enhancement of GABA-activated currents (EC(50), 0.4 microM in cultures; 1.4 microM in slices) and prolonged the duration of spontaneous and miniature inhibitory postsynaptic currents. Androstenol (0.1-1 microM) also potentiated the amplitude of GABA-activated currents in human embryonic kidney 293 cells transfected with recombinant alpha1beta2gamma2 and alpha2beta2gamma2 GABA(A) receptors and, at high concentrations (10-300 microM), directly activated currents in these cells. Systemic administration of androstenol (30-50 mg/kg) caused anxiolytic-like effects in mice in the open-field test and elevated zero-maze and antidepressant-like effects in the forced swim test (5-10 mg/kg). Androstenol, but not its 3beta-epimer, conferred seizure protection in the 6-Hz electroshock and pentylenetetrazol models (ED(50) values, 21.9 and 48.9 mg/kg, respectively). The various actions of androstenol in the whole-animal models are consistent with its activity as a GABA(A) receptor modulator. GABA(A) receptors could represent a target for androstenol as a pheromone, for which it is well suited because of high volatility and lipophilicity, or as a conventional hormonal neurosteroid.

Abstract


Each morning for a month female subjects placed either 5 alpha-androst-16-en-3 alpha-ol, a putative human pheromone, or a placebo on the upper lip. Each evening the subjects rated on five scales their moods during that day. In the middle of their monthly cycle those females exposed to androstenol rather than a control tended to rate their moods as submissive rather than aggressive. The compound did not significantly influence ratings of being happy/depressed; lethargic/lively; sexy/unsexy; irritable/good-tempered. The results are discussed in terms of the possible increased olfactory sensitivity of the human female to androstenol in the middle of her monthly cycle.
